Therapeutic Category
Neuropathic Pain Management & Neurotropic Supplementation
Product Overview
MACBA MD3 combines pregabalin, an α2δ ligand used in the management of neuropathic pain, with methylcobalamin and vitamin D3 to provide complementary neurotropic and nutritional support.
Clinical Pharmacology
Pregabalin
Pregabalin binds to the α2δ subunit of voltage-gated calcium channels in the central nervous system, reducing the release of excitatory neurotransmitters and thereby reducing abnormal neuronal excitability associated with neuropathic pain.
Methylcobalamin
Methylcobalamin is an active form of vitamin B12 involved in neuronal metabolism and maintenance of normal neurological function.
Vitamin D3
Vitamin D3 contributes to calcium-phosphate homeostasis and normal neuromuscular function.
Indications & Usage
MACBA MD3 is indicated for the management of neuropathic pain, including:
- Diabetic peripheral neuropathic pain
- Peripheral neuropathic pain
- Other neuropathic pain conditions where pregabalin therapy is clinically appropriate
Contraindications
Known hypersensitivity to pregabalin or any component of the formulation.
Warnings & Precautions
Dizziness and somnolence: Pregabalin may cause dizziness and drowsiness. Patients should exercise caution when driving or operating machinery.
Renal impairment: Pregabalin is primarily eliminated through renal excretion. Dose adjustment is required according to renal function.
Withdrawal: Abrupt discontinuation should be avoided. Pregabalin should generally be tapered gradually under medical supervision.
Respiratory depression: Caution is required in patients with respiratory risk factors and those receiving concomitant CNS depressants.
Drug Interactions
Pregabalin may have additive CNS-depressant effects with:
- Opioids
- Alcohol
- Sedatives
- Hypnotics
- Other CNS depressants
Pregnancy & Lactation
Pregabalin should be used during pregnancy only when clearly indicated and after assessment of the potential benefits and risks.
Pregabalin is excreted in human milk. The decision to continue treatment during breastfeeding should be made by the treating physician.
Dosage & Administration
The dosage should be individualized according to the indication, therapeutic response and renal function.
Typical adult starting regimen: 75 mg twice daily, where clinically appropriate.
Dose adjustment may be required according to patient response and renal function.
Do not discontinue abruptly.
Adverse Reactions
Commonly reported adverse reactions include:
- Dizziness
- Somnolence
- Headache
- Fatigue
- Dry mouth
- Blurred vision
- Peripheral edema
- Weight gain
